The short answer
CBD has not been shown to increase eszopiclone blood levels in an exact-pair human study. Review co-use because CBD can cause drowsiness, but do not assume every CBD product acts like a strong CYP3A4 inhibitor.
What this answer covers
Oral cannabidiol products; prescription purified CBD, high-dose CBD/THC extracts and retail formulations are distinct exposures.
- Supplement or preparation
- Cannabidiol
- Medication ingredient
- eszopiclone
The proposed blood-level effect is unconfirmed
Eszopiclone is metabolized through CYP3A4 and CYP2E1. Its exposure increased with the strong inhibitor ketoconazole, but this does not establish CBD’s effect or an automatic CBD-related dose adjustment.
Human CBD probe studies differ
An acute high-dose CBD-plus-THC brownie increased exposure to midazolam, a CYP3A probe, in healthy adults. It had no CBD-only arm. Current EPIDIOLEX labeling reports unchanged midazolam concentrations with repeated purified cannabidiol. Neither result measured eszopiclone.
Drowsiness is a separate concern
Prescription cannabidiol can cause somnolence, and LUNESTA warns about combining CNS depressants. That supports a precaution about alertness without proving increased eszopiclone absorption or blood levels.
Make changes with the prescriber
Show the exact CBD and THC content and dosing schedule before adding or stopping the product. No protective spacing interval was established. Avoid driving while impaired. Stop LUNESTA and contact the prescriber promptly if complex sleep behaviors occur.
Evidence and practical considerations
Absorption & effectiveness
No exact CBD-eszopiclone human PK evidence. Acute CBD+THC midazolam exposure increase and repeated purified-CBD midazolam null prevent a uniform prediction; ketoconazole results cannot classify CBD products or prove increased eszopiclone absorption.
- Exact scope
- Oral cannabidiol products; prescription purified CBD, high-dose CBD/THC extracts and retail formulations are distinct exposures. with Eszopiclone461016 IN; oral LUNESTA tablets, distinct from racemic zopiclone.
- Medication form and route
- oral medication; oral supplement
What remains uncertain
- No unmeasured pair incidence, blanket class transfer or arbitrary spacing interval.
Factors that may matter: Preparation; Dose; Other medicines; Organ function.

Population: People prescribed eszopiclone
CYP3A4/2E1 metabolism; ketoconazole increased exposure2.2fold. Rifampicin induction evidence measured racemic zopiclone, extrapolated by label. Additive CNS depression and boxed complex sleep behavior warning.
How this applies: Exact eszopiclone RxCUI 461016 IN, with stated salt/formulation/route bridge.
Other-drug results do not establish exact botanical effects; single-dose lorazepam/paroxetine nulls do not predict complete absence of chronic pharmacodynamic effects.

Population: 18 healthy adults in randomized crossover study.
CBD+THC increased oral midazolam AUC56%; THC alone did not inhibit the tested CYP pathways.
How this applies: 18 healthy adults; no CBD-only arm, acute high oral dose and probe cocktail. Not a eszopiclone trial, and midazolam is not interchangeable with eszopiclone.

Population: People prescribed cannabidiol
Somnolence/sedation warning; cannabidiol750mg twice daily did not change midazolam plasma concentrations after a single oral midazolam2.5mg dose.
How this applies: Original source interpreted for eszopiclone only within the stated preparation and endpoint limits.
Prescription purified cannabidiol oral solution, not every retail CBD extract. Midazolam PK null is contrary probe evidence, not proof of no eszopiclone interaction.

Side effects & toxicity
CBD-related drowsiness may add to eszopiclone impairment even without a demonstrated pharmacokinetic interaction.
- Exact scope
- Oral cannabidiol; exact product and co-ingredients matter. with Eszopiclone oral tablets.
- Medication form and route
- oral medication; oral supplement
What remains uncertain
- No direct pair rate or demonstrated blood-level increase.
Factors that may matter: Dose; THC content; Other sedatives.
