What the combination trial found
In 25 severely depressed patients, a tryptophan-nicotinamide-imipramine regimen was compared with other treatments. No substantial treatment difference emerged, and higher tryptophan levels correlated with worse response in the combined group.
Why those research doses are not a guide
Nicotinamide was included to alter tryptophan breakdown. The small study does not establish a safe supplement dose with your prescription or show that more precursor improves response.
